Growth conditions: anaerobic 37 °C
Equipment needed: Autoclave Hungate tubes Gassing station
Compound Amount Unit Conc. [g/L] Conc. [mM]
dehydrated RCM medium 38.0 g 38 -
Sodium resazurin
(0.1% w/v)
0.5 ml 5.0e-4 0.002
Sodium carbonate 1.0 g 1 -
Distilled water 1000.0 ml - -
1 Dissolve dehydrated RCM medium (available from BD BBL, OXOID or MERCK) in distilled water and add 0.5 ml/l Na-resazurin solution (0.1% w/v), bring to the boil and cool to room temperature while sparging with 80% N2 and 20% CO2 gas mixture. Dispense medium under same gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave. Before use, add 1.0 g of sodium carbonate per liter medium from a sterile anoxic stock solution prepared under 80% N2 and 20% CO2 gas mixture. Check pH of medium after autoclaving and adjust pH to 6.8, if necessary.
