1 Adjust pH to 7.0 using H2SO4.
2 Prepare the medium under H2 gas and fill 20 ml medium into 100 ml serum bottles and seal with a rubber stopper. Steam medium for 3 hours on each of 3 successive days. Give an atmosphere of H2/CO2/O2 (78/20/2) with an overpressure of two atmospheres. Incubate under shaking at 65°C.
3 For heterotrophic growth add yeast extract 1.0 g, peptone 5.0 g and starch 2.0 g to above medium. Incubate at 65°C in gas phase of 80% N2 and 20% air.
Solution occurs in 24 other media (first defined in Medium 700).
