1 Sparge solution A with 100% N2 gas for 30 - 45 min to make it anoxic. Dispense solution under same gas atmosphere into anoxic Hungate-type tubes or serum vials and sterilize by autoclaving. Solutions B and D are autoclaved separately under 100% N2 gas atmosphere. Solution C is autoclaved under 80% N2 and 20% CO2 gas atmosphere. The medium is completed before use by adding appropriate amounts of solutions B, C and D to solution A. Adjust pH of complete medium to 7.2, if necessary. |
