1 Autoclave. After cooling to 50C and add aseptically 135 �l of conc. HCl (final conc. 0.05%). Then add aseptically the following solutions.
Fetal bovine serum
(Biowest; heat inactivated)
20.0 ml - -
Vitox
(Oxoid) (1 vial/10 ml of rehydration fluid)
2.0 ml - -
Skirrow supplement
(Oxoid) (1 vial/2 ml of sterile, purified water)
0.4 ml - -
Amphotericin B
(5 mg/ml DMSO)
100.0 �l - -
2 For stable culture, incubate the microorganisms in 5 ml of above-mentioned broth in a polystyrene culture flask (25 cmz2 flask, canted neck, vent cap, AS ONE: Cat. No. 2-8589-01) at 37C under a 5% O2 and 12% CO2 atmosphere (with shaking at low speed; 50 rpm).
