1 After preparing the Trypticase soy agar plate with 5%sheep blood, spreading the subculture on the agar surface and place one NAM disk aseptically. Incubate the agar plate under a H2-CO2-N2 (1:1:8) gas mixture. To prepare the NAM disk, impregnate with 20 �l filter-sterilized 1.5% NAM (N-acetylmuramic acid, Sigma) aqueous solution to a sterile paper disk (0.64 cm in diameter). Stored the air-dried disk at room temperature. |
