1 To complete medium, aseptically add the following solutions to the medium (per 10 ml):
Solution B 0.1 ml - -
Sodium acetate
(filter-sterilized, 1.0 M)
0.2 ml - -
NaHCO3
(filter-sterilized, 5%)
0.4 ml - -
2 Mix ingredients and adjust pH to 7.2. Distribute the solution into tubes (e.g., 10 ml in the Hungate tubes) under a N2-CO2 gas mixture (4:1, v/v), seal with butyl rubber stoppers and autoclave.
