1 Prepare the liquid medium. Final pH 5.5.The medium is fairly weakly buffered so the pH should be checked before and after autoclaving. Dispense the medium into serum bottles. Add 10-20% methane to the gas phase. Sterilize by autoclaving.
2 The strain should be grown with shaking.
