1. Adjust solution A for final pH 6.0 with HCl. Autoclave solution A at 121°C for 15 min. Cool to room temperature and aseptically add solution B filter sterilised.

0.2	g	Fish-Sperm DNA	
0.02	g	Phenol red	
0.4	g	Urea	
700	ml	Water	


## Solution B

100	ml	Yeast extract (BD, 2%)	
200	ml	Horse serum	
5	ml	Isovitalex
