# 1205: 1/2 TRYPTIC SOY BROTH (ANAEROBIC)

13.75	g	Trypticase soy broth (BD BACTO)	
1	g	Yeast extract	
2.5	g	NaCl	
3.6	g	D-Glucose	
0.15	g	DL-Dithiothreitol (DTT)	
1000	ml	Distilled water	

1. Dissolve ingredients except glucose and DTT, then sparge medium with 100% N2 gas for 30 - 45 min to make it anoxic. Adjust pH to 7.2 with HCl, then dispense medium under same gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave at 121°C for 20 min. Add glucose and DTT from anoxic stock solutions prepared under 100% N2 gas atmosphere and sterilized by filtration.
