Add to 1000 ml of medium after autoclaving: 1 ml **Trace element solution SL-10** 1 ml **Vitamin solution** 2. adjust pH to 5.5 3. For solid medium use purified agar (Oxoid LP0028) ## SSE (double concentrated) 0.2938 g CaCl2 x 2 H2O 0.1069 g NH4Cl 0.2036 g MgCl2 x 6 H2O 0.1983 g (NH4)2SO4 0.739 g MgSO4 x 7 H2O 0.8606 g CaSO4 x 2 H2O 0.236 g Ca(NO3)2 x 4 H2O 0.424 g Na NO3 0.5 ml KH2PO4 (100 mM solution, 100 mM) 0.0111 g FeSO4 x 7 H2O 0.087 g K2SO4 1000 ml Distilled water 1. Dissolve by shaking overnight. Sterilize by autoclaving. ## Trace element solution SL-10 10 ml HCl (25%) 1.5 g FeCl2 x 4 H2O 70 mg ZnCl2 100 mg MnCl2 x 4 H2O 6 mg H3BO3 190 mg CoCl2 x 6 H2O 2 mg CuCl2 x 2 H2O 24 mg NiCl2 x 6 H2O 36 mg Na2MoO4 x 2 H2O 990 ml Distilled water 1. First dissolve FeCl2 in the HCl, then dilute in water, add and dissolve the other salts.
