# 388: DICTYOGLOMUS MEDIUM ## Main sol. 388 1.5 g KH2PO4 4.2 g Na2HPO4 x 12 H2O 0.5 g NH4Cl 0.38 g MgCl2 x 6 H2O 0.06 g CaCl2 x 2 H2O 0.04 g Fe(NH4)2(SO4)2 x 6 H2O 2.9 ml CoCl2 x 6 H2O (0.1% w/v) 2.4 ml Na2MoO4 x 2 H2O (0.1% w/v) 1.7 ml Na2SeO3 x 5 H2O (0.01% w/v) 2 ml MnCl2 x 4 H2O (0.1% w/v) 2.8 ml ZnSO4 (0.1% w/v) 2 g Yeast extract 2 g Polypeptone 5 g Starch (soluble) 0.5 ml Sodium resazurin (0.1% w/v) 1 g Na2CO3 1 g L-Cysteine HCl x H2O 1 ml **Wolin's vitamin solution (10x)** 1000 ml Distilled water 1. Dissolve ingredients (except carbonate, cysteine and vitamins), bring medium to the boil, then cool to room temperature under 100% N2 gas atmosphere. Add and dissolve carbonate and cysteine, adjust pH to 7.2, then dispense under 100% N2 gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave. Add vitamins from an anoxic stock solution prepared under 100% N2 gas and sterilized by filtration.
