25	g	Tryptone	
7.5	g	Yeast extract	
20	ml	Tris-HCl (pH 7.5, 1M)	
1000	ml	Distilled water	

1. Autoclave, cool, and then add: 

5	ml	MgCl2 (1M)	
10	ml	Diaminopimelic acid (1%)	
10	ml	Thymidine (0.4%)	
25	ml	Glucose (20%)	

2. Magnesium chloride should be sterilized separately by autoclaving, and the other ingredients should be sterilized separately by filtration.
