# 411: CLOSTRIDIUM ACETOBUTYLICUM MEDIUM

200	g	Potato (fresh, washed, peeled and sliced)	
2	g	CaCO3	
0.5	ml	Sodium resazurin (0.1% w/v)	
6	g	D-Glucose	
0.5	g	L-Cysteine HCl x H2O	
1000	ml	Distilled water	

1. Mix potatoes and CaCO3 then autoclave 30 min at 121°C. Filter through cheese cloth, add glucose and cysteine, distribute under 100% N2 gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave at 121°C for 20 min. Adjust pH of complete medium to 6.8.
2. Note: For the reactivation of freeze-dried strains it is recommended to use the following simple milk medium:

100	ml	Milk (fresh)	
0.1	ml	Sodium resazurin (0.1% w/v)	

1. Adjust pH to 7.1. Tube and autoclave under 100% N2 gas atmosphere for 12 min at 121°C.
