# 48: TRYPTONE THIOGLYCOLATE MEDIUM ## Main sol. 48 5.45 g K2HPO4 1.2 g KH2PO4 25 mg MgSO4 x 7 H2O 15 mg CaCl2 x 2 H2O 10 mg FeSO4 x 7 H2O 2 mg MnCl2 x 4 H2O 2.5 mg CoCl2 x 6 H2O 2.5 mg Na2MoO4 x 2 H2O 20 g Glucose 2 g Peptone 2 g Tryptone 6 g Yeast extract 0.5 g Na-thioglycolate 1000 ml Distilled water 1. Dissolve ingredients (except glucose), adjust pH to 7.5 and sparge medium with 100% N2 gas for 30 - 45 min to make it anoxic. Dispense medium under same gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave. 2. Add glucose to the medium after autoclaving from a sterile stock solution prepared under 100% N2 gas.
