1. Rehydrate and grow lyophilized cells from the ampoule in 5 ml liquid broth 545. Subsequent subculturing may be carried out in liquid medium 545 or on agar plates.


## 545. TRYPTONE SOYA BROTH (TSB)

17	g	Casein peptone	
3	g	Soy peptone	
2.5	g	D(+)-Glucose	
5	g	NaCl	
2.5	g	K2HPO4	
15	g	Agar	
1000	ml	Distilled water	

1. Adjust pH to 7.3.
2. Medium 545 is the same as Caso Bouillon (Merck or Oxoid, Germany).

30	g	Trypticase soy broth (BBL 11768, Oxoid CM129 or Merck 5459)	
20	g	Agar	
1000	ml	Distilled water	

1. pH 7.3
2. Autoclave at 121°C for 15 min.
