# 796: ARCHAEOGLOBUS VENEFICUS MEDIUM ## Main sol. 796 18 g NaCl 0.33 g KCl 7.15 g MgCl2 x 6 H2O 0.25 g NH4Cl 0.14 g CaCl2 x 2 H2O 0.18 g K2HPO4 x 3 H2O 1 ml **Trace element solution** 2 ml Fe(NH4)2(SO4)2 x 6 H2O (0.1% w/v) 0.5 ml Sodium resazurin (0.1% w/v) 4 g NaHCO3 1 g Na2SO3 1 g Na-acetate 0.5 g Na2S x 9 H2O 1000 ml Distilled water 1. Dissolve ingredients (except bicarbonate, sulfite, acetate and sulfide), then sparge medium with 80% N2 and 20% CO2 gas mixture for 30 - 45 min to make it anoxic. Thereafter, add and dissolve bicarbonate and equilibrate the medium with the 80% N2 and 20% CO2 gas mixture to achieve a medium pH of 7.0. Dispense medium under same gas atmosphere into serum vials (e.g., 25 ml per 100 ml bottle) and autoclave. After sterilization add sulfite, acetate and sulfide from sterile anoxic stock solutions prepared under 100% N2 gas. The stock solution of sulfite should be freshly prepared and sterilized by filtration. Prior to inoculation check pH of complete medium and adjust to 7.0, if necessary. 2. After inoculation, pressurize bottles to 1 bar overpressure with sterile 80% H2 and 20% CO2 gas mixture. ## Trace element solution 5.8 g MnCl2 x 4 H2O 10 g NaCl 2 g (NH4)2Ni(SO4)2 1 g FeSO4 x 7 H2O 1 g CoCl2 x 6 H2O 1 g CaCl2 x 2 H2O 1 g ZnSO4 x 7 H2O 0.1 g CuSO4 x 5 H2O 0.1 g H3BO3 0.1 g AlK(SO4)2 x 12 H2O 0.1 g Na2MoO4 x 2 H2O 0.1 g Na2WO4 x 2 H2O 0.1 g Na2SeO3 x 5 H2O 1000 ml Distilled water 1. Adjust pH to 1.0 with HCl.
