# 939: METHYLOTROPHIC ARTHROBACTER AND HYPHOMICROBIUM MEDIUM ## Main sol. 939 7.9 g Na2HPO4 x 2 H2O 1.5 g KH2PO4 0.8 g NH4Cl 0.1 g MgSO4 x 7 H2O 10 ml **Trace elements solution** 1000 ml Distilled water 10 ml Methanol 1. Final pH 7.2-7.5 2. After autoclaving add 10 ml methanol to 1 litre of the sterile Mineral Salts Solution. ## Trace elements solution 50 g EDTA 1 g ZnSO4 x 7 H2O 7.34 g CaCl2 x 2 H2O 2.5 g MnCl2 x 4 H2O 0.5 g CoCl2 x 6 H2O 0.5 g (NH4)6Mo7O24 5 g FeSO4 x 7 H2O 0.2 g CuSO4 x 5 H2O 1000 ml Distilled water 1. Dissolve the EDTA in about 400 ml of water, then add 9 g NaOH. Dissolve each of the salts individually in about 40-50 ml of water and add them to the EDTA-NaOH solution. Adjust the final pH of the solution to pH 6.0 with 1M NaOH (about 24 ml). Make up to 1 litre with distilled water, store in the dark and do not autoclave the stock solution before it is added to the medium.
