# J1082: HALALKALIARCHAEUM MEDIUM


## Main sol. J1082

937.5	ml	Basal mineral NaCl medium (sterilized, see Medium No. 1079)	
62.5	ml	**Soda based mineral medium** (sterilized)	

1. To complete the medium, aseptically add the following solution (autoclaved or *filter-sterilized): 

5	ml	Sodium pyruvate (*, 2 M)	
10	ml	Trace vitamins (see Medium No. 197) (*)	
0.2	ml	Yeast extract (10%)	


## Soda based mineral medium

185	g	Na2CO3	
35	g	NaHCO3	
16	g	NaCl	
1	g	K2HPO4	
5	g	KCl	

1. Add components to distilled water and bring volume to 1.0 L. Autoclave and let it stand for three days. Decant the clear solution into another empty sterile bottle. Aseptically add per liter the following solutions (autoclaved): 

1	ml	NH4Cl (4 M)	
1	ml	**Selenite-tungstate solution**	
1	ml	**Trace element solution**	
1	ml	MgCl2 (1 M)
