1 Mix ingredients, except Na2S x 9H2O, adjust pH to 6.0 with H2SO4. Sterilize by filtration while gassing with H2-CO2 (80:20) gas mixture. Autoclave Na2S x 9H2O as a 5% solution under a N2 atmosphere. Prior to inoculation, anaerobically add the sterile Na2S x 9H2O solution, and readjust pH to 6.5 with sterile H2SO4. Pressurize the inoculated bottle to 200 kPa H2-CO2 (80:20).
